Antagonization of monoamine reuptake transporters by agmatine improves anxiolytic and locomotive behaviors commensurate with fluoxetine and methylphenidate

Abstract Background Agmatine (AGM) is known for its protective effects including neuroprotection, nephroprotection, gastroprotection, cardioprotection, and glucoprotection. Studies have validated the neuroprotective role of AGM as antidepressant, anxiolytic, locomotive, antipsychotic agent in psychopathologies. Fluoxetine (FLX) most extensively prescribed antidepressant while methylphenidate (MPD) frequently psychoactive stimulant ADHD (attention deficit hyperactivity disorder) treatment worldwide. The mechanism action FLX MPD involves reuptake inhibition serotonin dopamine norepinephrine at presynaptic transporters. Present study was designed to determine safety efficacy administration along with conventional psychostimulative drugs. also aimed establish underlying monoamine Results significantly ameliorated locomotion activity box open field anxiolytic behaviors light/dark transition EPM were improved ( p <0.01). growth appetite animals enhanced antidepressive behavior FST Moreover, co-administration or rats’ compared single administration. Conclusion determined significant locomotor, MPD. showed rats treated combined doses food intake body weights. This has proposed potential receptors that may lead transporters increase 5-HT, D, NE concentrations synaptic level.
